本发明提供了一种DNA点突变定量检测方法,通过PNA探针两次对野生型模板的抑制,极大程度地抑制了野生型模板的扩增,解决了PNA钳制不完全的问题。同时,利用扩增引物Tm值差异和扩增时退火温度控制,使巢式PCR能在单管中进行,增强了扩增强度和扩增特异性。使拷贝数极低的临床样本的扩增成为了可能。最后,通过实时荧光的闭管检测,既实现了实时检测,又避免了样品的交叉污染,还实现了突变的定量。此外,该方法只需要普通实时荧光PCR仪器,且在2 h内就能得到检测结果,试剂成本低,可实现对样品的快速、低成本检测。总之,该方法的成功建立,可为临床DNA点突变的检测提供一种高灵敏、高特异、简便快速、无污染和低成本的检测新方法。
